SewLow Discount Fabrics in East Cambridge (on Cambridge Street) and SewFisticated (in the Twin Cities Plaza near East Cambridge) are the two I go to the most often for fabric. The Singer store in Davis Square on Elm St is useful for supplies like thread, needles, and interfacing but doesn't really have much in the way of fabric.
There's a sewing store in Davis near the Starbucks. It's not that great. You'd be better off taking the Red Line down to Downtown Crossing and going to Chinatown to Winmil Fabrics on 111 Chauncy St. There's another good fabric store on the same street.
Otherwise, some of my friends out in the suburbs go to Jo-Ann fabrics and I believe that there's a decent store in the Twin Cities Plaza in Cambridge near Union Square, Inman Square, and Lechmere.
